Iconic how this film gave the Lupin series this iconic vehicle which would be used in later material for this franchise.

Technically it was Part 1 of the Lupin Series to first feature the 500 (/vehicle_938266-Fiat-500-F-110F-1965.html), but it was Cagliostro that made it really popular

According to Miyazaki, "While Lupin III's cars became increasingly extravagant with each series, Lupin III himself is someone who sympathizes with the poor, a poor man's son. Therefore, he always had to drive a Fiat Nuova 500."
When the film was screened in Italy, Miyazaki was told that he could have received advertising fees from Fiat if he had declared it in advance. However, Miyazaki responded by saying, "If I had known it would be screened in Italy, I would have chosen a different manufacturer." This was because he believed that audiences would enjoy the film more if the vehicles featured were unfamiliar to them.
Incidentally, at the time of the film's theatrical release, Yasuo Otsuka owned a car of the same model (FIAT NUOVA 500) that appeared in the film.

Yasuo Outsuka (the Character Design and Animation Director) owned an Fiat 500 during the production of Cagliostro

The first Fiat 500 was the first one imported to Japan.
Yasuo Otsuka, known in the animation industry and among fans as an unparalleled car enthusiast, became a Fiat owner for the first time more than 55 years ago.
"When I was younger, I drove a certain Japanese car. One day, my regular salesman told me, 'We're going to start handling the Fiat 500,' and I reserved one before I even saw the actual car (laughs). What sealed the deal for me was its cute design and the fact that the roof could be opened. And also, the fact that it had a rear engine, which I loved." Then, Mr. Otsuka made a surprising statement: "According to the salesman, the Fiat 500 I first drove was the very first one imported into Japan. That means I was the first owner of a Fiat 500 in Japan." Mr. Otsuka says, "Of all the cars I've driven, the Fiat 500 was the most fun!" He was so captivated by the charm of the Fiat 500 that he owned three of them in succession: white, cream, and light blue.

Driving around Japan with the Fiat 500 "I had so much fun with the Fiat 500," says Ms. Otsuka with a smile. Traveling around Japan in the Fiat 500 is a fun memory she still cherishes, and the most memorable trip was the one to Kyushu. "At that time, the highway system wasn't fully developed yet, so I drove all the way from my home in Tokyo to Kyushu on local roads."
The key points in choosing a car, as depicted in the artwork:
Mr. Otsuka commuted to his workplace in his beloved Fiat 500. The Fiat 500 that appears in the masterpiece "Lupin the Third: The Castle of Cagliostro" was apparently drawn in an environment where his beloved car was always by his side.
"I had parked my Fiat 500 in a spot visible from the studio window, so all the staff members were sketching my beloved car while looking at it. At the time, my 500 was a light blue color. As for why we made Lupin III's 500 yellow... I guess it was just a spur-of-the-moment idea (laughs)."
